New York residents and regular visitors question

I could ask elsewhere, but this forum has the highest proportion of Americans than any other I inhabit

I land at JFK sometime Saturday and I'm staying a block or two west of Times Square. We had a minibus transfer included last time, but this time we thought we'd wing it.

What's the best way to get from JFK to central Manhattan in the middle of the day, Town Car/Yellow Cab/something else ?
